is it possible to assign two keys to one button for CoD4

Not yet. This is a heavily requested feature and should be added sometime in the future.

Perhaps you could provide more detail in what you are trying to do. If you want two keys to be pressed at the same time then with the Assignment window open (looks like a keyboard) click the Advanced button on the lower right side of the window. Now you may add as many keys as you like. When you press the button on the controller all keys will be sent at the same time. If you want the keys to be sent one after another then add pauses between keys, now when you press the button the keys are sent in order. If you want the button to send each key one at a time then add cycle stops between the keys. Now one key will be sent for each press of the controller button. If you want one key to be pressed when you tap the button and a different key if you hold the button then that is not possible like mnl said but may be added in the future.
